RESUMO

Oxaliplatin is an important initial chemotherapy benefiting advanced-stage colorectal cancer patients. Frustratingly, acquired oxaliplatin resistance always occurs after sequential chemotherapy with diverse antineoplastic drugs. Therefore, an exploration of the mechanism of oxaliplatin resistance formation in-depth is urgently needed. We generated oxaliplatin-resistant colorectal cancer models by four representative compounds, and RNA-seq revealed that oxaliplatin resistance was mainly the result of cells' response to stimulus. Moreover, we proved persistent stimulus-induced endoplasmic reticulum stress (ERs) and associated cellular senescence were the core causes of oxaliplatin resistance. In addition, we screened diverse phytochemicals for ER inhibitors in silico, identifying inositol hexaphosphate (IP6), whose strong binding was confirmed by surface plasmon resonance. Finally, we confirmed the ability of IP6 to reverse colorectal cancer chemoresistance and investigated the mechanism of IP6 in the inhibition of diphthamide modification of eukaryotic elongation factor 2 (eEF2) and PERK activation. Our study demonstrated that oxaliplatin resistance contributed to cell senescence induced by persistently activated PERK and diphthamide modification of eEF2 levels, which were specifically reversed by combination therapy with IP6.

RESUMO

Excessive levels of cadmium (Cd) in soil exert serious negative impacts on soil ecosystems. Microorganisms are a common component of soil and show great potential for mitigating soil Cd. This review summarizes the application and remediation mechanisms of microorganisms, microbial-plants, and microbial-biochar in Cd-contaminated soil. Microorganisms such as Bacillus, Acinetobacter, Pseudomonas, and arbuscular mycorrhizal fungi (AMF) can change the biological validity of Cd through adsorption, mineralization, precipitation and dissolution. Different factors such as pH, temperature, biomass, concentration, and duration have significant effects on Cd bioavailability by microorganisms. Pseudomonas, Burkholderia, and Flavobacterium can promote the uptake of Cd2+ by hyperaccumulator through promotion and activation. Biochar, a soil amendment, possesses unique physicochemical properties and could act as a shelter for microorganisms in agriculture. The use of combined microbial-biochar can further stabilize Cd compared to using biochar alone.

RESUMO

This study aimed to explore the predictive factors of nonmalignant pathological diagnosis and final diagnosis of ultrasound-guided cutting biopsy for peripheral pulmonary diseases. A total of 470 patients with peripheral lung disease diagnosed as nonmalignant by ultrasound-guided cutting biopsy in the First Affiliated Hospital of Guangxi Medical University from January 2017 to May 2020 were included. Ultrasound biopsy was performed to determine the correctness of pathological diagnosis. Independent risk factors of malignant tumor were predicted by multivariate logistic regression analysis. Pathological biopsy results showed that 162 (34.47%) of the 470 biopsy data were specifically benign, and 308 (65.53%; malignant lesions: 25.3%, benign lesions: 74.7%) were nondiagnostic findings. The final diagnoses were benign in 387 cases and malignant in 83 cases. In the nondiagnostic biopsy malignant risk prediction analysis, lesion size (OR = 1.025, P = 0.005), partial solid lesions (OR = 2.321, P = 0.035), insufficiency (OR = 6.837, P < 0.001), and presence of typical cells (OR = 34.421, P = 0.001) are the final important independent risk factors for malignant tumors. In addition, 30.1% (25/83) of patients with nonmalignant lesions who were finally diagnosed with malignant tumors underwent repeated biopsy, and 92.0% (23/25) were diagnosed during the second repeated biopsy. 59.0% (49/83) received additional invasive examination. Nondiagnostic biopsy predictors of malignant risk include lesion size, partial solid lesions, insufficiency, and presence of atypical cells. When a nonmalignant result is obtained for the first time, the size of the lesion, whether the lesion is subsolid, and the type of pathology obtained should be reviewed.

RESUMO

Cadmium-contaminated soils are an urgent problem that needs to be solved in many countries and regions. In this study, a new heavy metal passivator, micro-nano nitrogen-doped biochar (Nm-NBC), was prepared by introducing nitrogen into biochar. Soybean was used as an experimental plant to compare the effects of corn straw biochar (CBC, not modified), ammonium chloride modified corn straw biochar (NBC), and micro-nano nitrogen-doped biochar (Nm-NBC) on the remediation of Cdcontaminated soil. The results showed that the biomass of soybean, pH, organic matter, and total nitrogen content of the Cd-contaminated soil significantly increased, and the available Cd in soil significantly reduced (P < 0.05) when CBC, NBC, and Nm-NBC were added. The effect was as follows: Nm-NBC > NBC > CBC; Nm-NBC had the best result. When 1% Nm-NBC added to the soil, the Cd content in beans reduced by 68.09%. BET, FTIR, XPS, and SEM were used to analyze the characteristics of Nm-NBC and its mechanisms in the remediation of Cd-contaminated soils. The results showed that Nm-NBC had larger specific surface area and abundant functional groups; -COOH and graphitic nitrogen in Nm-NBC can form Cd-O bond and Cd-π with Cd(II) in the soil. Therefore, Nm-NBC prepared by introducing nitrogen into biochar has a promising application in the remediation of Cd-contaminated soil.

RESUMO

OBJECTIVES: This is a prospective study to evaluate the clinical value of high-frequency ultrasound (HFUS), superb microvascular imaging (SMI), and contrast-enhanced ultrasound (CEUS) in differentiation of pigmented villonodular synovitis (PVNS) and highly active rheumatoid arthritis (RA). METHODS: Twenty PVNS patients and 24 active RA patients were selected to undergo HFUS, SMI, and CEUS examinations. The characteristics of HFUS, SMI, and CEUS in PVNS and RA were compared, and the differential diagnosis performances of HFUS, SMI, and CEUS in PVNS and RA were evaluated by receiver operating characteristic (ROC) analysis. RESULTS: There were statistically significant in joint effusion, synovial thickness, synovial morphology, synovial echo, synovial vessel shape, synovial enhanced direction, and enhanced pattern between PVNS and RA (P < .05). However, no statistically significant were found in bone erosion, synovial boundary, blood signal grading of synovium, synovial enhanced strength, and CEUS quantitative parameters (including PI, TTP, S, MTT, and AUC) (P > .05). The AUC of HFUS, SMI, and CEUS for differential diagnosis PVNS and RA were 0.832, 0.675, and 0.817, respectively. The AUC of HFUS + SMI, HFUS + CEUS, SMI + CEUS, HFUS + SMI + CEUS were 0.923, 0.940, 0.817, and 0.940, respectively. The AUC of HFUS + SMI and HFUS + CEUS was higher than that of each alone (P < .05). CONCLUSIONS: HFUS, SMI, and CEUS can be used as supplementary methods for diagnosis and differential diagnosis in PVNS and active RA. What is more, the combination of HFUS + SMI and HFUS + CEUS was suggested.

RESUMO

Background: Some studies found that family doctor contract services (FDCSs) had positive impact on the self-measurement behaviors of hypertension patients. However, evidence concerning the association between FDCSs and blood pressure measurement awareness among hypertension patients is not clear. Objective: This study aims to explore the relationship between FDCSs and blood pressure measurement awareness among the hypertension patients, and examine whether there is a difference in this relationship among middle-aged and aged adults in rural Shandong, China. Methods: A multi-stage stratified random sampling was adopted in 2018 in Shandong Province to conduct a questionnaire survey among the sample residents, in which 982 hypertension patients were included in the study. Pearson chi-square test and logistic regression model were employed using SPSS 24.0 to explore the association between FDCSs and blood pressure measurement awareness. Results: 76.8% of hypertension patients would measure blood pressure regularly. The blood pressure measurement awareness of the signing group was significantly higher than that of the non-signing group when controlling other variables (P < 0.001, OR = 2.075, 95% CI 1.391-3.095). The interaction of age and contracting status were significantly correlated with blood pressure measurement awareness (P = 0.042, OR = 1.747, 95% CI 1.020-2.992; P = 0.019, OR = 2.060, 95% CI 1.129-3.759). Factors including gender (P = 0.011, OR = 0.499, 95% CI 0.291-0.855), household income (P = 0.031, OR = 1.764, 95% CI 1.052-2.956), smoking status (P = 0.002, OR = 0.439, 95% CI 0.260-0.739), sports habits (P < 0.001, OR = 2.338, 95% CI 1.679-3.257), self-reported health (P = 0.031, OR = 1.608, 95% CI 1.043-2.477), distance to the village clinic (P = 0.006, OR = 1.952, 95% CI 1.208-3.153) and medications (P < 0.001, OR = 3.345, 95% CI 2.282-4.904) were also found to be associated with the blood pressure measurement awareness of hypertension patients. Conclusion: The government should take efforts to strengthen publicity and education of family doctors and pay more attention to uncontracted, middle-aged, female patients and patients with unhealthy life behaviors to improve the blood pressure measurement awareness.

RESUMO

BACKGROUND: To evaluate the diagnostic performance of ultrasound-guided percutaneous needle biopsy (US-PNB) for peripheral pulmonary lesions on the basis of the intention-to-diagnose principle and to identify the risk factors for diagnostic failure. METHODS: Patients with peripheral pulmonary lesions who underwent US-PNB in the Department of Ultrasound, the First Affiliated Hospital of Guangxi Medical University, China, from January 2017 to December 2019 were retrospectively enrolled in this study. The diagnostic accuracy, sensitivity, specificity, positive predictive value (PPV), and negative predictive value (NPV) of ultrasound-guided biopsy in peripheral pulmonary lesions was assessed. The proportion of diagnostic failure was calculated and the associated risk factors were identified. RESULTS: A total of 930 patients were enrolled in this study, including 957 US-PNBs. The overall diagnostic accuracy, sensitivity, and specificity was 90.3% [864/957; 95% confidence interval (CI): 0.88-0.922], 87.2% (495/568; 95% CI: 0.84-0.90), and 94.9% (369/389; 95% CI: 0.92-0.97), respectively. The PPVs and NPVs were 99.4% (495/498; 95% CI: 0.98-0.99) and 88.1% (369/419; 95% CI: 0.85-0.91), respectively. The diagnostic failure rate was 9.72% (93/957; 95% CI: 0.08-0.12). The independent risk factors for diagnostic failure were history of smoking [odds ratio (OR) 1.78, 95% CI: 1.13-2.78], 18G biopsy needle (OR 2.97, 95% CI: 1.06-8.30), and final clinical diagnosis of malignant lesions (OR 2.72, 95% CI: 1.63-4.55). CONCLUSIONS: US-PNB of peripheral pulmonary lesions show high diagnostic efficacy. A history of smoking, use of 18G biopsy needle, and final clinical diagnosis of malignant lesions are independent risk factors for diagnostic failure of US-PNB in peripheral pulmonary lesions. Identification of the independent risk factors for diagnostic failure can help reduce the diagnostic failure rate.

RESUMO

Considering that hyperaccumulators can accumulate high concentrations of iron salt, they can successfully obtain magnetic hydrochar from iron-rich hyperaccumulators. In this study, iron-rich biomass was obtained by irrigating Phytolacca acinosa Roxb. using iron salt. Magnetic nano-Fe3O4 hydrochar was prepared from iron-rich Phytolacca acinosa Roxb. via hydrothermal carbonization to remove Cd. The characterization results showed that the synthesized magnetic nanoparticles had an average size of 2.62 ± 0.56 nm and N elements were doped into magnetic nano-Fe3O4 hydrochar with abundant oxygenic groups. Cd adsorption on magnetic nano-Fe3O4 hydrochar was better fitted using the Langmuir isotherm and the pseudo-second-order kinetic model. The maximum adsorption capacity was 246.6 mg g-1 of Cd. The research confirmed that Cd adsorption was controlled by multiple mechanisms from the jar test, transmission electron microscopy mapping, scanning electron microscopy-energy dispersive X-ray spectroscopy, X-ray diffraction, Fourier transform infrared spectroscopy, and X-ray photoelectron spectroscopy. CdCO3 crystals can be formed after adsorption, indicating that surface precipitation played an important role in Cd adsorption. The abundance of O atoms and the doping of N atoms on the hydrochar surface were conducive to Cd adsorption, indicating that the mechanisms were related to surface complexation and electrostatic attraction. In addition, the significant decrease in Na+ content after Cd adsorption illustrated that ion exchange had a non-negligible effect on Cd adsorption. This study not only provides a strategy for preparing magnetic nano-Fe3O4 hydrochar derived from iron-rich plants but also verifies multiple Cd adsorption mechanisms using magnetic nano-Fe3O4 hydrochar.

RESUMO

Anthropogenic activities have caused extensive arsenic (As) contamination in soils. The role of biochar in the remediation of As-contaminated soils has been attracting attention lately. In this study, effects of straw biochar, iron oxide, and iron oxide-modified biochar on soil microbial community composition and soil chemical properties were tested in an As-contaminated soil. After 9 months of incubation, soil chemical properties and microbial communities were analyzed. Our results showed that biochar addition significantly increased soil pH value, soil organic carbon (SOC) concentration, and the ratio of soil carbon to nitrogen (soil C:N ratio) but decreased soil dissolved organic C. Adding iron oxide also increased soil pH value, while iron oxide-modified biochar decreased it. Interestingly, compared with the control, all treatments significantly decreased soil total microbial biomass and biomasses of soil bacteria, fungi, Actinomyces, and protozoa. In addition, significantly positive correlations were found between soil pH and soil total microbial biomass as well as bacterial, Actinomyces, and arbuscular mycorrhizal fungal biomass. There were negative relationships between SOC, soil C:N ratio, and all soil microbial biomass indicators in all treatments. These results indicated that biochar and iron oxide-modified biochar affected soil microbial community composition by altering the soil C:N ratio, but iron oxide affected it via adjusting soil pH. Furthermore, the iron oxide-modified biochar effects on soil microbial community and soil chemical properties are not the same as the additive effects of biochar and iron oxide alone, and its effect on soil microbial community is regulated by the soil C:N ratio. These findings will help guide the development of remediation practices for As-contaminated soil using biochar.

RESUMO

BACKGROUND AND OBJECTIVES: To evaluate the feasibility and efficacy of single-fiber laser ablation (LA) under ultrasound guidance and appropriate ablation modes in the treatment of selected metastatic lymph nodes of papillary thyroid carcinoma (PTC) and benign cold thyroid nodules STUDY DESIGN/MATERIALS AND METHODS: A total of 18 patients (consisting of 8 patients with 18 metastatic lymph nodes of PTC and 10 patients with 10 benign cold thyroid nodules) each underwent one session of single-fiber LA under ultrasound guidance. On the basis of the sizes of the nodules, the ablation modes were chosen accordingly. The single-dot ablation mode was used in the nodules with three orthogonal diameters measuring no greater than 10 mm in diameter, with a dot, a level and an insertion. The double-dots overlapping ablation mode was used in the nodules with the largest diameters measuring greater than 10 mm (in which the nodules measured no more than 15 mm in diameter and with the other two perpendicular diameters measuring no greater than 10 mm in diameter) with two dots, a level and two insertions. The multiple levels and dots overlapping ablation mode was used in the nodules with the three orthogonal diameters all measuring larger than 10 mm, with multiple dots, levels and insertions. RESULTS: After 12 months of follow-up in the treated nodules of the metastatic lymph nodes of PTC and benign cold thyroid nodules, the mean baseline volumes decreased from 0.29 ± 0.12 to 0.03 ± 0.03 ml and 3.85 ± 0.64 to 1.1 ± 0.37 ml, respectively, and the mean volume reduction ratios (VRRs), which was calculated as {[(initial volume-final volume) × 100%]/initial volume}, were 90.3 ± 7.6% and 72 ± 5.8%, respectively. There were six ablative zones that completely disappeared, whereas the ablative zones that still existed presented as scar-like areas or small hyperechoic areas that were compatible with scar tissue among the 18 malignant nodules at the last follow-up. CONCLUSIONS: Single-fiber LA under ultrasound guidance, with the appropriate ablation modes, is feasible and effective for the treatment of selected metastatic lymph nodes of PTC and benign cold thyroid nodules. This study suggests that single-fiber LA may be applied to selected cases with appropriate ablation modes. Lasers Surg. Med. 2019 © 2019 Wiley Periodicals, Inc.

RESUMO

High-power tunable pulsed and CW mid-infrared fiber gas laser sources in acetylene-filled hollow-core fibers, to the best of our knowledge, are demonstrated for the first time. By precisely tuning the wavelength of the pump source, an amplified tunable 1.5 µm diode laser, to match different absorption lines of acetylene, the laser output is step-tunable in the range of 3.09~3.21 µm with a maximum pulse average power of ~0.3 W (~0.6 µJ pulse energy) and a maximum CW power of ~0.77 W, making this system the first watt-level tunable fiber gas laser operating at mid-infrared range. The output spectral and power characteristics are systemically studied, and the explanations about the change of the ratio of the P over R branch emission lines with the pump power and the gas pressure are given, which is useful for the investigations of mid-infrared fiber gas lasers.

RESUMO

We report here the first watt-level efficient single-pass 1.54 µm fiber gas Raman source by methane-filled hollow-core fiber operating at atmospheric pressure. Pumped with a high-power MOPA (master oscillator power amplifier) structure Q-switched 1.06 µm pulsed solid-state laser, efficient 1.54 µm Stokes wave is generated in a single-pass configuration by vibrational stimulated Raman scattering of methane molecules. With an experimentally optimized fiber length of 3.2 m, we get a 1543.9 nm Stokes wave operating at atmospheric pressure with a record average power of ~0.83 W, which is about 12 times higher than the similar experiment previously reported, and the corresponding power conversion efficiency is about 45%. Operating at atmospheric pressure makes it more convenient in future applications.

RESUMO

A retrospective study was carried out to evaluate the diagnostic accuracy and the factors influencing the diagnostic accuracy of 648 procedures of ultrasound-guided percutaneous needle biopsy (PNB) for peripheral pulmonary lesions (PPLs). We reviewed the histopathology results, the clinical records and the procedure reports of these 648 biopsies and the final diagnoses of 637 PPLs to determine the diagnostic accuracy of ultrasound-guided PNB. Factors that influenced the diagnostic accuracy were assessed by analysis of the biopsy procedures, which were classified as diagnostic cases (true-positive and true-negative) and non-diagnostic cases (false-positive, false-negative and indeterminate). Statistical analyses of factors that related to patient demographic characteristics, lesion characteristics and biopsy details were performed to determine possible effects on diagnostic accuracy. Biopsies were successfully performed in all cases, and 11 patients underwent second biopsies for the same lesions. Among the 637 PPLs, there were 326 (51.2%) malignant lesions, 272 (42.7%) benign lesions and 39 (6.1%) indeterminate lesions. Of the 272 benign lesions, 114 (41.9%) were found to be tuberculous. The overall diagnostic accuracy was 81.8%, and the rates of hemoptysis, symptomatic pneumothorax and chest-tube insertion were 8.0%, 1.7% and 0.9%, respectively. Lesions sizes were divided into 3 groups according to the measurement by ultrasound. For lesions that measured ≤20 mm, 21-49 mm and ≥50 mm, the diagnostic accuracy was 72.0%, 86.8% and 79.7%, while sensitivity and specificity were 54.3%-79.2%, 88.3%-90.7% and 79.4%-89.5% and 77.3%-100%, 96.8%-100% and 58.6%-100%, respectively. Diagnostic accuracy was significantly affected by lesion size when lesion size was measured by ultrasound (p = 0.006) and computed tomography (CT) (p = 0.001). In the 3 lesion groups of ≤20 mm, 21-49 mm or ≥50 mm, diagnostic accuracy among each group was significantly different (p <0.001). When lesion size was measured by ultrasound (p <0.001) and CT (p <0.001) and the 3 groups were analyzed (p <0.001), there was a statistically significant relationship between lesion size and the presence of necrosis. The rates of the presence of necrosis in lesions that measured ≤20 mm, 21-49 mm and ≥50 mm were 3.9%, 11.7% and 28.8%, respectively. No significance was found for age (p = 0.119), gender (p = 0.25), lesion location (p = 0.55), the presence of necrosis (p = 0.226), patient position (p = 0.25), needle size (p = 0.26), puncture angle (p = 0.34) and needle passes (p = 0.21). Ultrasound-guided PNB is an effective and safe diagnostic method for PPLs; the diagnostic accuracy is significantly affected by lesion size and decreases in smaller (≤20 mm) and larger (≥50 mm) lesions.

RESUMO

Herein, biochar derived from lotus seedpods, as an effective adsorbent, was prepared by pyrolysis method at 300 and 600 °C. The physicochemical characteristics and cadmium adsorption properties were studied systematically by batch adsorption experiments, FTIR, SEM-EDX, XRD, and XPS. Cd adsorption onto lotus seedpod-derived biochar was better fitted using Freundlich isotherm and pseudo-second-order model. Adsorption capacity of biochar produced at 300 and 600 °C was 31.69 and 51.18 mg g-1, respectively. The Cd adsorption capacity of biochar was related to its characteristics determined by pyrolysis temperature, including carbonization, surface area, surface morphology, and surface functional groups. Cd adsorption on lotus seedpod-derived biochar revealed that adsorption was controlled by multiple mechanisms including surface complexation, ion exchange, surface precipitation, and Cd-π interaction. This study showed that lotus seedpod-derived biochar is an effective and environmentally friendly adsorbent for water treatment.

RESUMO

We report here for the first time, to the best of our knowledge, a 100-kW-peak-power, GHz-linewidth, sub-nanosecond, 1.9-µm laser source by gas stimulated Raman scattering (SRS) in hollow-core fiber. A H2-filled, anti-resonance, hollow-core fiber is pumped with a sub-nanosecond, high-peak-power, 1064-nm microchip laser, generating a 1908-nm Stokes wave by vibrational SRS of H2 molecules. A maximum peak power of about 150 kW (average power 55 mW, pulse energy 55 µJ) is achieved with a 1.4-m fiber length and only 3 bar H2 pressure. The maximum quantum efficiency is about 54%, and the corresponding slope efficiency is about 37%. The linewidth of the Stokes wave is about 2 GHz, which decreases about 1-2 orders compared with the rare-earth-doped fiber lasers of the same peak-power level. Operation close to atmospheric pressure makes it more convenient in future applications. If a tunable pump laser is used, a high-peak-power, narrow-linewidth, broadly tunable, 2-µm fiber laser source can be easily achieved.

RESUMO

To evaluate acoustic radiation force impulse (ARFI) inthe differential diagnosis of small (≤20 mm) solid breast lesions and identify the most efficient ARFI parameters. Conventional ultrasonography and ARFIwere performed in 120 patients with 121 small solid breast lesions. The area ratios (ARs) of the lesion on virtual touch tissue compared to B-mode were calculated. The shear wave velocity of the inner (SWVi) and boundary (SWVb) of the lesions and surrounding fatty tissue (SWVf) was measured. The ratio of SWVi to SWVf (SWVrat) was calculated. AR, SWVi, SWVb, and SWVrat were significantly larger in malignant lesions (all P < 0.001). A cutoff AR of 1.17 yielded the highest area under the receiver operating characteristic curveamong the various parameters (91.2% sensitivity, 85.9% specificity, 88.4% accuracy) for the differential diagnosis of small breast lesions, but this value did not significantly differ from SWVi (P = 0.1144). This AR cutoff indowngradingcategory 4a to category 3 would avoid 83.3% unnecessary biopsies, and improved diagnostic specificity up to 73.4% without decreasing sensitivity. AR and SWVi are efficient parameters for the differential diagnosis of small breast lesions, whichwill improve diagnostic specificity and reduce unnecessary biopsies.

RESUMO

Nano-drug delivery systems that integrate inorganic and organic or even bioactive components into a single nanoscale platform are playing a hugely important role in cancer treatment. In this article, the fabrication of a versatile nanocarrier based on self-assembled structures of gold nanoparticles (AuNPs)-zein is reported, which displays high drug-loading efficiency for needle-shaped hydroxycamptothecin (HCPT) nanocrystals. The surface modification with folate-conjugated polydopamine (PFA) renders them stable and also facilitates their selective cellular internalization and enhancement of endocytosis. The release of payloads from nanocomplexes (NCs) was shown to be limited at physiological pH (17.1±2.8%) but significantly elevated at endosomal/lysosomal pH (58.4±3.0%) and at enzymatic environment (81.4±4.2%). Compared to free HCPT and its non-targeting equivalent, HCPT@AuNPs-Zein-PFA exerted a superior tumor suppression capacity as well as low side effects due to its active and passive targeting delivery both in vitro and in vivo. These results suggest that the NCs with well-defined core@shell nanostructures encapsulated with HCPT nanocrystals hold great promise to improve cancer therapy with high efficiency in the clinic. STATEMENT OF SIGNIFICANCE: A novel nanocomplex with HCPT nanocrystals encapsulated was designed to achieve selective cellular uptake by endocytosis, acid responsive release in the tumor microenvironment and excellent tumor suppression without toxicity. This nanocomplex with conjugation of folate was stable in the bloodstream, with minimal drug release in extracellular conditions, leading to prolonged blood circulation and high accumulation in tumor tissues. The entrapment of a nanocrystal drug into nanomaterials might be capable of delivering drugs in a predictable and controllable manner.

RESUMO

OBJECTIVE: To retrospectively analyze and compare the ultrasonographic characteristics and BI-RADS-US classification between patients with BRCA1 mutation-associated breast cancer and those without BRCA1 gene mutation in Guangxi, China. METHODS: The study was performed in 36 lesions from 34 BRCA1 mutation-associated breast cancer patients. A total of 422 lesions from 422 breast cancer patients without BRCA1 mutations served as control group. The comparison of the ultrasonographic features and BI-RADS-US classification between two the groups were reviewed. RESULTS: More complex inner echo was disclosed in BRCA1 mutation-associated breast cancer patients (x(2) = 4.741, P = 0.029). The BI-RADS classification of BRCA1 mutation-associated breast cancer was lower (U = 6094.0, P = 0.022). CONCLUSIONS: BRCA1 mutation-associated breast cancer frequently displays as microlobulated margin and complex echo. It also shows more benign characteristics in morphology, and the BI-RADS classification is prone to be underestimated.

RESUMO

The continuous changes in pulmonary hemodynamic properties and right ventricular (RV) function in pulmonary arterial hypertension (PAH) have not been fully characterized in large animal model of PAH induced by a carotid artery-jugular vein shunt. A minipig model of PAH was induced by a surgical anastomosis between the left common carotid artery and the left jugular vein. The model was validated by catheter examination and pathologic analyses, and the hemodynamic features and right-ventricle functional characteristics of the model were continuously observed by Doppler echocardiography. Of the 45 minipigs who received the surgery, 27 survived and were validated as models of PAH, reflected by mean pulmonary artery pressure ≥25 mmHg, and typical pathologic changes of pulmonary arterial remodeling and RV fibrosis. Non-invasive indices of pulmonary hemodynamics (pulmonary artery accelerating time and its ratio to RV ventricular ejection time) were temporarily increased, then reduced later, similar to changes in tricuspid annular displacement. The Tei index of the RV was elevated, indicating a progressive impairment in RV function. Surgical anastomosis between carotid artery and jugular vein in a minipig is effective to establish PAH, and non-invasive hemodynamic and right-ventricle functional indices measured by Doppler echocardiography may be used as early indicators of PAH.
